The most common neurodegenerative disorders—AD and PD—share several clinical features. Although different hypotheses regarding the cause of these diseases have been advanced, progressive cell death and the underlying mechanisms are not yet known. Abnormal immune-related phenomena have been postulated as one of the mechanisms underlying the pathogenesis of both disorders. The evidence for such mechanisms include the presence of autoantibodies against dopaminergic and cholinergic neurons, a decreased immunoglobulin secretory capacity, and the alteration of cytokine production in the brain and peripheral blood cells of patients with PD. In addition, alterations in the content of several cytokines and growth factors in the brain parenchyma of patients with AD and PD are well documented.1,2⇓ Recently, we and others have found that the risk for developing AD is considerably increased in carriers of a polymorphism of the interleukin-1α gene (−889; IL-1A).3,4⇓
